Machine PrecisionTm = [ ΔH · 1000 / (ΔS + R · ln(C/4)) ] - 273.15 + 16.6 · log₁₀[Na_eq] | Ta = min(Tm) - 5°C
Step-by-Step Mathematical Derivation
Melting temperature is determined by enthalpy (ΔH) and entropy (ΔS) transitions between adjacent base pairs: Tm = ΔH / (ΔS + R·ln(C/4)).
Mg²⁺ ions stabilize DNA phosphate backbones. Owczarzy et al. (2008) salt equivalence: [Na⁺_eq] = [Na⁺] + 120 × √([Mg²⁺] - [dNTP]).
Optimal primer extension occurs at Ta = Tm - 5°C. For efficient PCR, the melting temperatures of the forward and reverse primers must match within ΔTm ≤ 2.0°C.
