Machine PrecisionRRF = (Area_imp / Conc_imp) / (Area_std / Conc_std) | % Impurity = [ (Area_sample_imp · Conc_std) / (Area_std · Conc_sample · RRF) ] · 100
Step-by-Step Mathematical Derivation
The detector response factor RF represents detector sensitivity (Peak Area / Concentration). Relative Response Factor is the sensitivity ratio: RRF = RF_impurity / RF_API.
If an impurity has a weak chromophore (RRF < 1.0), it absorbs less UV light per mole than the API. Using raw area percent underestimates the impurity. Dividing by RRF scales it up to true mass balance.
ICH Q3A/B mandates that any RRF outside 0.8 to 1.2 MUST be explicitly applied. If RRF is between 0.9 and 1.1, a factor of 1.0 is sometimes acceptable.
